Human CD4+ T Helper Cell Responses after Tick-Borne Encephalitis Vaccination and Infection. A DNA immunization model study with constructs expressing the tick-borne encephalitis virus envelope protein E in different physical forms. Altogether, the findings suggest that noninfectious VLPs can serve as a potentially safe and effective vaccine candidate against diverse subtypes of TBEV.Īberle, J.H., Aberle, S.W., Allison, S.L., Stiasny, K., Ecker, M., Mandl, C.W., Berger, R.,Heinz, F.X., 1999. Immunization with the VLP vaccine induced the production of multiple-cytokine-producing antiviral CD4+ T cells in vivo, including TNF-α+, IL-2+, and IFN-γ+ T cells. Furthermore, the group that received the VLP vaccine did not exhibit significant pathological changes and the inflammatory factors were significantly suppressed compared to the control group. The VLPs provided protection to mice lacking the type I interferon receptor (IFNAR) against lethal TBEV challenge, with undetectable viral load in brain and intestinal tissues. ![]() These findings indicated that the VLP-based vaccine elicited the production of cross-subtype reactive antibodies. The efficacy of the VLPs was subsequently evaluated in C57BL/6 mice, and the resultant IgG serum could neutralize both Far-Eastern and European subtypes of TBEV. The present study describes a novel strategy for the assembly of virus-like particles (VLPs) by co-expressing the structural (core/prM/E) and non-structural (NS2B/NS3Pro) proteins of TBEV. The coverage and immunogenicity of the currently available vaccines against TBEV are relatively low therefore, it is crucial to develop novel and effective vaccines against TBEV. ![]() ![]() Tick-borne encephalitis virus (TBEV) is an important tick-borne pathogen that poses as a serious public health concern. ![]() ![]() setup ( Hands On Throttle And Stick) which in my opinion is a much more versatile and immersive. In part this is because I have a wide range of interests and skills, but this is not the only reason. You’ll notice on my checklist that there’s a pretty wide variety of activities represented. The combination of answers I’m presented with gives me better questions to ask when it comes time to negotiate. It helps me know where they’re at-are they new to the activity, do they have notes about whether it’s something they only do with people they have particular chemistry with… you get the idea. I ask them to rate: by experience level-never tried, tried but not enough to fully evaluate, or experienced by how much they enjoy the activity-0-5 by whether or not the activity is a limit, a curiosity of theirs, or a valued part of play for them to let me know if the activity is a fetish or something they feel they want to be “forced” to do in order to get over their nerves to try and finally I offer space for notes and questions.Īll of this information gives me a much clearer picture of how my partner feels about an activity, than if they’d simply said, “I rate such-and-such activity as a 3”. In my checklist I ask people to rate each activity in a number of different ways in order to get a more comprehensive idea about how my sub actually experiences each activity. It’s important to remember though that people interpret different terms differently, and one person’s idea of what “medium masochism” is might be wholly different than another’s.Īlongside the long list of activities to be rated, there are often a few ways each activity can be rated. This is so the person filling it out can do so as clearly as possible, and the person reading it can interpret their answers relatively accurately. How to use a BDSM checklist: At the top of most comprehensive checklists you’ll find definitions about what words mean and instructions about how to fill the pages out. Feel free to download, edit, update, change, and utilize the list for yourself: It’s not an exhaustive list in the least bit, but I think it’s a pretty good start and it works for me rather well. ![]() My list is based off a few I’ve used in the past, with various bits taken off, added on, recategorized, redefined, and with slightly different options than I’ve found on some. I’ll try to update the file link when I think of it in the future. That’s just the nature of this ever-evolving beast. I know I’ll be refining it as long as I use it. Today I’m sharing a BDSM checklist that I’ve put together. Certainly it’s impossible to develop an exhaustive list of things to try, but even still some lists cater more towards styles of play I enjoy, and some are far from useful to me specifically. There are some I find to be too complicated, and others too simplistic. Sometimes I get annoyed that a checklist’s language is different from what I’m used to, or the list comes off as too heteronormative or less than gender inclusive. You can find a bunch of them online, and over the years I’ve found examples which work more or less to my satisfaction. A helpful tool I frequently use with new subs and trainees is the BDSM checklist. ![]() ![]() ![]() However, virtually no literature exists on the usefulness of the ecomap with non-Western cultures. Helping professionals from the United States, Canada, the United Kingdom, and New Zealand have written about the value of utilizing family diagrammatic tools with families. Two such resources are "Ecotivity" by Wonderware and Mattaini's software companion to "Visual Ecoscan for Clinical Practice." In recent years, new computer software allows professional helpers to develop computer-generated diagrammatic assessments such as ecomaps and genograms. Although originating in social work, the use of this tool spans disciplinary boundaries literature on nursing, psychology, law, and other disciplines salutes the usefulness of family diagrammatic tools. Other social workers who have joined Hartman in refining the ecomap include Joan Laird and Mark Mattaini. Since its creation, the ecomap has been used in a variety of practice settings. ![]() The concept quickly became popular with family therapists in the United States and the United Kingdom. The social worker Ann Hartman first introduced the ecomap in 1978 in her article "Diagrammatic Assessment of Family Relations." Hartman's work evolves from the school of family theories known as family systems theory, which grew out of the general systems theory applied to sciences such as physics, biology, and anthropology. This empowers families to know where to begin making changes. Using the ecomap, the therapist and family can identify the external relationships that are nourishing, as well as those that are wounded. Arrows indicated direction of energy flow and conflicted or broken relationships may be represented by interrupted lines. For example, a solid line may indicate a strong and healthy relationship, while a diffused line represents a weaker tie. Standard symbols are used to express energy that flows from a person or family to other important people and institutions. ![]() The ecomap diagram consists of circles, lines, and arrows (see Figure 1). Identifying these connections clarifies and organizes data on a family's environment highlights energy that flows into and out of the family and raises issues such as network size and stability, reciprocity of relationships, and access to or deprivation of available resources. As the name signifies, therapist and client together map out connections between the family and its ecological environment. The ecomap, also called a sociogram, is a visual assessment tool depicting the relationships between a family and its social network. ![]() ![]() ![]() A typical use of the NATO Phonetic Alphabet would be to spell out each letter in a word over the phone by saying, for example: "S as in Sierra" (or "S for Sierra"), "E as in Echo, Y as in Yankee, F as in Foxtrot, R as in Romeo, I as in India, E as in Echo, D as in Delta" to communicate the spelling of the name "Seyfried" correctly. These are used to avoid misunderstanding due to difficult to spell words, different pronunciations or poor line communication. ![]() ![]() One last thing… Without Extensions like Artisan, Vertex Tools, SubD, etc I personally would not be using SketchUp. Why because if not I was afraid it would stop being developed.Īnyways, I really hope this tool is a monetary success for the devs because I want it to be further developed in the future and not have to stop because of a lack of resources. Else they will go extinct, that is why I was more than ok for SketchUp itself to move to the subscription model. If you look at other software then you will see almost all of them are moving to subscription. Now I dedicate less time to developing my tools which are going to be available as a subscription as well by the way. Instead, I have to use my time to work on jobs not related to SketchUp so I can support myself. I myself have had to stop developing plugins full-time because I was not able to make enough money selling them. I have seen this before on an Animation Plugin named ‘SimFonIA Animation Tools’ which was great but the developers had to stop because of lack of resources I suspect. The REAL problem I see is that developers would STOP developing SketchUp extensions because of a lack of resources and support from users not wanting to spend money on their work. $8.25 a month for Artisan is less than what you spend on food each day. So if you look at it that way subscription is not that different. For example, we went from Vertex Tools to Vertex Tools 2. What normally happens with the “Perpetual” license is that at the end of the year, the extension releases a new version. It is NOT fair for developers to dedicate many years working for us to only pay just $30 to $40 bucks. When it comes to software it makes sense it is a subscription because the developers constantly work on the code and customer support. ![]() Supports 3 axes modes (Local, View, World).New Tool-based UI to easily select the plane using inferencing.New Scale Tool UI similar to native SketchUp scale. ![]() Added automatic quad auto-folding and repair to maintain quads during transform operations.Added ability to transform FFD control points.Added ability to lock axis inference using arrow keys.Added 3 axes modes (Local, View, World) - use ALT to switch mode.Added ALT modifier to control whether hidden vertices are excluded from window selection.Added support for FFD control point selection.Added support for edge loop selection via double-click.Simply click the Symmetry button to create a mirrored component for symmetrical sculpting. Subtract the number of turns across the back dowel from those on the front dowel to find the difference between them. Since the front is wider you must add more turns on its left and right sides so the center of the pattern will run straight from back to front. Using the number of turns on the front and back dowels, you need to (a) account for width difference between front and back and (b) determine how many groups of 4 cords you will make.Īccount for width difference. Lay out the "front-to-back" pattern (yellow). Try to wrap evenly and snuggly for best results. It's worth the time to quickly wrap each side and get a good count. I tried to estimate this by wrapping a few inches and then multiplying by the length (pic 2). In essence of time, I suggest wrapping around and around the entire length of the back dowel, front dowel and one side to get an accurate count of turns. ![]() You can always add more cord on the bottom side so it's not critical to be exact.įirst find out how many turns you'll be making on each side. This is a good estimate for similar sized chairs. ![]() The yellow & black chair measured 10.5" on each side, 13" front and 11" back and required 110 ft of yellow cord and 165 ft of black cord. The only tricky part of this project is spacing the weave pattern evenly although it's pretty easy. ![]() ![]() ![]()
